Hypoxia level of 2D/3D-cultured cells was evaluated by using Cell3iMager duos.

Materials and Methods

  • Cell Line: HepG2 cells (RIKEN BRC)
  • Medium: DMEM (Nacalai tesque)
  • Plate for 2D culture: 96-well plate flat bottom (Corning)
  • Plate for 3D culture: 96-well plate flat bottom ULA (Corning)
  • Seeding cell density: 0 - 2000 cells/well
  • Culture days: 4 days
  • Reagent: Hypoxia probe LOX-1(Organogenix)
  • Imaging methods: Fluorescence, Low magnification lens

Cell3iMager duos is very useful for cell counting.
It is possible to analysis for various fluorescent regents and other cells.

Results and Conclusions

  • These figures show that Hypoxia level in 3D-cultured cells is bigger than in 2D-cultured cells.
  • Hypoxia level was calculated from red intensity sum.
