Objective

The cell density that stepwise cell number seeding, was quantyfied by using Cell3iMager duos.

Materials and Methods

  • Cell Line:
    • HeLa cells (ATCC)
    • NIH3T3 cells (RIKEN BRC)
  • Medium: DMEM (Nacalai tesque)
  • Reagent:
    • Hoechst33342 (Nacalai tesque)
    • Calcein-AM (Nacalai tesque)
  • Plate:
    • 96-well plate flat bottom (Sumitomo Bakelite)
    • 96-well half area plate flat bottom (Corning)
  • Seeding cell density: 0 - 2000 cells/well
  • Culture days: 3 - 4 days
  • Imaging methods:
    • fluorescence field (exitation; Blue and UV)
    • High magnification lens

Results and Conclusions

  • Cell3iMager duos is very useful for cell counting.
  • It is possible to analysis for various fluorescent regents and other cells.
